Adolescent idiopathic scoliosis (AIS) exerts significant adverse effects on the physical and mental health of numerous adolescents. However, the underlying mechanisms of AIS remains controversial. This study is conducted to determine the potential diagnostic value of miR-183-5p in AIS and to preliminarily explore its regulatory mechanisms. This study detected the expression level of miR-183-5p in the serum of AIS patients by reverse transcription quantitative real‑time polymerase chain reaction (RT-qPCR), and receiver operating characteristic (ROC) curves were constructed to evaluate its diagnostic potential. Human bone marrow mesenchymal stem cells (hBMSCs) were induced to undergo osteogenic differentiation. Cell proliferation and apoptosis were assessed using cell counting Kit-8 (CCK8) assay and flow cytometry, respectively. The target genes of miR-183-5p were predicted using online databases. The relationship was verified using the dual-luciferase reporter gene assay and Spearman’s correlation analysis. The circulating miR-183-5p was upregulated in AIS patients and had a good diagnostic value. Overexpression of miR-183-5p suppressed the expression of osteogenic differentiation markers and cell viability. The target gene of miR-183-5p was GNG12, and a negative correlation was observed between miR-183-5p and GNG12. Upregulation of GNG12 reversed the inhibitory effect induced by upregulated miR-183-5p in hBMSCs. Overexpressed miR-183-5p has diagnostic potential for AIS. It inhibits osteogenic differentiation and cell proliferation by negatively regulating GNG12, and this regulatory axis may be associated with the pathogenesis of AIS.
