The study aimed to systematize information and critically contextualize the history and challenges of vaccine development, focusing primarily on serious adverse reactions in light of the COVID-19 pandemic and the ChAdOx1 nCov-19 immunobiological. To this end, an integrative literature review was conducted, outlined by a search for scientific articles and bibliographies across major national and international electronic repositories. The descriptors used for the search were: SARS-CoV-2, COVID-19, history, immunization, vaccines, vaccine development, ChAdOx1 nCov-19, BNT162b2, adverse effects, and thrombosis, as well as their respective English translations. The findings externalize and reinforce that such occurrences do not compromise the efficacy and safety of vaccines; rather, they drive technological refinement, the improvement of surveillance, and a deeper understanding of immune-mediated mechanisms. Consequently, the development of immunizers and new public health strategies are strengthened, contributing to the control and prevention of infectious diseases.
