This method demonstrates monofluoromethylation of benzils and isatins using radical chemistry, suggesting new pathways for synthesizing bioactive molecules.
A new method to synthesize α-CFH2 carbinols has been developed through the photoredox-catalyzed direct radical hydromonofluoromethylation of benzils/isatins, 9H-fluoren-9-one, 2-methylisoindoline-1,3-dione, etc., using bench-stable H2CFSO2Na. It was further applied to the synthesis of a bioactive isatin-derived molecule. Mechanistic studies reveal that radical-radical coupling between the •CFH2 radical and the benzoin radical affords the final monofluoromethylated products.
