This review summarizes various fzd7 inhibitors and their role in targeting wnt receptors in tumors, suggesting future strategic directions.
Frizzled-7 (FZD7) is a key Wnt receptor that is frequently overexpressed in aggressive solid tumours and fuels therapy resistance. Over the past several years, a diverse arsenal of FZD7 inhibitors, including monoclonal antibodies, antibodydrug conjugates, bifunctional fusion proteins, small-molecule and peptideshas been discovered or synthesized by researchers, and it has been proven that tumor cell amplification or resuscitation can be controlled by inhibiting FZD7. Among them, Humanised antibody SHH002-hu1, the latest antibody targeting FZD7, achieves high affinity with low cross-reactivity. Yet no agent has progressed beyond phase Ib. After analyzing the latest researches, this article synthesise the current landscape, classifying inhibitors by modality, then provides a comprehensive reference for the new drug design of the target and suggests the future use direction of FZD7 inhibitors. In the future, the selectivity of targeted FZD7 antibodies may become the focus. At the same time, the combined drug use strategy of the target inhibitor with other anticancer drugs and the individualized medication method for different patients will also become the direction of research.
