Background: Age estimation & sex determination in both adults and children become most important identifying features in case of legal issues, unknown individuals, deaths and mass disaster. The use of non-destructive methods like exfoliative cytology and liquid based cytology enhances the scope of non-invasive approach for age estimation, sex determination in forensic odontology. Aim & Objectives: “To evaluate the efcacy of Romanowsky stain (Leishman Giemsa Cocktail Stain) & Papanicolaou (PAP) stain in conventional and liquid based oral exfoliative cytology smears and to correlate Nuclear area (NA), Cell area (CA) and N/C ratio for age estimation and sex determination using morphometric analysis in study groups. 400 smears from 100 app ”Materials & Method: “ arently healthy individuals were prepared by conventional and liquid based cytology followed by staining with LG Cocktail stain and PAP stain. Morphometric analysis for NA, CA and N/C ratio was done accompanied by correlation with age and sex of individuals. Correlation ”Results: “ of mean cell area, nuclear area and N/C ratio in liquid based cytosmear and conventional cytosmear shows statistically signicant results. There is Signicant decrease in nuclear area and N/C ratio from age group <20 years to older age group and signicant variations in cellular and nuclear parameters among different study groups. LG Cocktail stain shows more efcient staining of cytoplasm and nucleus with comparable cellular and nuclear morphological details with PAP stain.”Conclusion: Our study results revealed signicant age and sex related variations in cell area, nuclear area, N/C ratio in oral exfoliated cells.
