French, and co-workers engineer blood–brain tumor barrier (BBTB) models via two-photon polymerization. 3D micro-porous capillary-like structures support endothelial alignment, cytoskeletal organization, and pericyte-endothelial-glioma tri-cultures, highlighting the role of glioma cells in barrier destabilization. The structures are also integrated within microfluidic chips, enabling stable perfusion and offering a versatile platform to study tumor-vascular interactions as well as barrier integrity in the context of glioma research.
