Purpose: Although medical treatment is preferred in the early acute phase of peyronie disease (PD), current literature on its effectiveness is insufficient. The aim of this study is to compare the effectiveness of oral colchicine and potassium para-aminobenzoate (PPA). Methods: Patients treated for PD in our clinic between 2012 and 2021 were reviewed retrospectively. Patients were treated with colchicine ( n = 28) or PPA ( n = 30) for 3 months. The effects of both drugs on the plaque size, plaque calcification, penile pain, and degree of penile curvature were compared between the groups. Results: Patients’ mean age was 52 ± 5.9 years; the median time of onset of symptoms was 4 months (range, 2–6 months). The PPA treatment significantly improved penile deviation (33.5%), penile plaque size (28.7%) and penile pain in addition prevented plaque calcification. The colchicine treatment also improved the penile deviation, penile plaque size, and penile pain; however, the comparison of pre- and post-treatment comparisons of these parameters were insignificant. PPA was more effective for penile deviation at 3 and 6 months. Although no significant difference was observed in the regression of penile pain at 3 months, PPA was found to be more effective than colchicine at 6 months. There was no difference in the effects on the plaque size between the treatments at 3 and 6 months. Conclusion: PPA is more effective than colchicine in regression of penile deviation, pain, and plaque calcification.
