To determine the expression profile and functional activity of Toll-like receptors across murine dendritic cell subtypes compared to human patterns.
Assessed mRNA expression profiles of Toll-like receptors across murine splenic dendritic cell subsets, including plasmacytoid and CD8α+ populations.
Evaluated functional activation by measuring ex vivo survival, surface marker upregulation, and IL-12p40 production following stimulation with a TLR7 ligand.
Murine dendritic cell subsets generally expressed similar levels of most Toll-like receptor mRNAs, but CD8α+ dendritic cells selectively lacked TLR5 and TLR7 while preferentially expressing TLR3.
TLR7 ligand exposure induced activation, survival, surface marker upregulation, and IL-12p40 production in CD8α- dendritic cells and plasmacytoid dendritic cells, but failed to activate CD8α+ dendritic cells.
