To determine the distribution, biophysical properties, and functional impact of hyperpolarization-activated inward currents (Ih) along the somatodendritic axis of hippocampal CA1 pyramidal neurons.
Obtained cell-attached patch-clamp recordings from soma and apical dendrites of hippocampal CA1 pyramidal neurons to characterize Ih density, kinetics, and conductance properties.
Conducted dual whole-cell patch-clamp recordings along the somatodendritic axis to assess regional input resistance, membrane time constants, and signal propagation.
Ih current density increased by more than sixfold from the soma to distal dendrites, exhibiting mixed Na+-K+ conductance and sensitivity to low concentrations of external CsCl.
Higher dendritic Ih density lowered local input resistance and accelerated membrane polarization rates, significantly decreasing EPSP amplitude and duration while restricting temporal summation.
Dendritic Ih exerted minor effects on backpropagating action potentials, primarily reducing spike duration and enhancing afterhyperpolarization to dampen subthreshold dendritic excitability.
