The in vitro synthesis of the aniline–deoxyguanosine adduct 1 was achieved both with pure deoxyguanosine and with DNA. N Acetoxyaniline, a potential metabolite and possibly the ultimate carcinogen of aniline, was employed for this purpose. It was synthesized from phenylhydroxylamine and AcCN and reacts with deoxyguanosine to give 1 in 6% yield.
