The (pro)renin receptor functions in both renin-angiotensin system-dependent and -independent pathways, and its blockers show potential in mitigating end-organ damage.
This review summarizes the molecular mechanisms of the (pro)renin receptor in both RAAS and Wnt signaling pathways, highlighting the therapeutic potential and current controversies of (P)RR blockers.
May guide (P)RR blocker development; leaves open clinical adoption pending randomized trials.
Pro)renin receptor [(P)RR] is one of the major components of the renin angiotensin (RA) system, which is involved in the regulation of blood pressure. (P)RR binds with and increases the activity of mature renin, the key regulatory enzyme of RA system, which catalyzes the conversion of angiotensinogen to angiotensin-I. (P)RR also binds to the inactive form of renin called prorenin.

Biswas et al. (2014) conducted a review in Renin-angiotensin system related diseases. (Pro)renin receptor blockers (e.g., Aliskiren, Decoy peptide) was evaluated. The (pro)renin receptor functions in both renin-angiotensin system-dependent and -independent pathways, and its blockers show potential in mitigating end-organ damage.
