Do large doses of warfarin sodium reduce ADP-induced platelet aggregation and impair haemostasis in vivo in rats?
Do large doses of warfarin sodium reduce ADP-induced platelet aggregation and impair haemostasis in vivo in rats?
In a rat model, supratherapeutic doses of warfarin significantly reduce ADP-induced platelet aggregation and impair haemostasis.
Does not support changes to clinical warfarin use; leaves open relevance of high-dose platelet effects in humans.
Large doses of warfarin that induced prothrombin levels considered unsafe for man significantly reduced ADP aggregation of platelets in vivo in the rat. The effectiveness of the platelet plug in haemostasis was also severely impaired. The possible mechanisms for this effect are discussed. A survey of the literature of the effect of coumarin therapy on experimental thrombosis is given.
