Presents a draft genome sequence and analyzes antibiotic resistance and virulence in Staphylococcus saprophyticus from cows with subclinical mastitis.
This study presents the draft genome sequence of Staphylococcus saprophyticus strain 3M isolated from a cow with subclinical mastitis (SCM) in Kiruhura district, Uganda and explores the isolates resistance and virulence potential. The assembled genome size was 2,791,445 bp with a 33% GC, 13 Contigs, Contig N50 2,589,240, Contig L50 1 and 2,791,440x Genome coverage. Check M analysis revealed 68.25% completeness. Average Nucleotide Identity (ANI) analysis revealed Staphylococcus saprophyticus subsp. saprophyticus ATCC 15,305 = NCTC 7292(GCA_006094355.1) as the closest type strain with an ANI of 99.52%, 87.49% assembly coverage, 93.95% type assembly coverage, confirming species-level identity and close relatedness. Screening with the Comprehensive Antibiotic Resistance Database (CARD) and the Virulence Factor Database (VFDB) revealed several genes conferring resistance to different classes of antibiotics: antibiotic tet (K)- Tetracyclines, norA, YkkCD, ANT (6)-I, str, and S12p- Aminoglycosides. And also, Virulence factors included: Agr- biofilm and toxins release, TcaR- Protein A, MprF- resist the host’s innate immune system (specifically, antimicrobial peptides). Other genome characteristics are presented in Table 1 below.
