This repository contains the research proposal for a pilot case-control study investigating the incidence and genetic risk contribution of the PDX1 E224K (c.670G>A) mutation in individuals with Type 2 Diabetes Mellitus (T2DM) in Sri Lanka. The proposal presents the study rationale, objectives, literature review, methodology, ethical considerations, and planned statistical analyses. The proposed research aims to determine the frequency of the PDX1 E224K mutation among Sri Lankan individuals with T2DM and healthy controls using genomic DNA extraction, polymerase chain reaction (PCR), Sanger sequencing, and sequence analysis. By evaluating the association between this genetic variant and T2DM susceptibility, the study seeks to generate preliminary evidence that may support future large-scale genetic studies and contribute to a better understanding of diabetes genetics within the Sri Lankan population.
