Topoisomerase assays have evolved from simple catalytic assays such as relaxation and supercoiling to those which help to determine the structure-activity relationships of inhibitory agents such as quinolones and some antitumor agents. Topoisomerase assays have found further use in the detection of enzyme fractions during the purification of topoisomerases as well as in studies of mechanisms of antibacterial, antiparasitic, and antitumor agents
