A new vitamin B12 compound has been recognized ionophoretically and isolated in small amounts from Propionibacterium shermanii. Its properties suggest that it is B12 5'-phosphate. The new compound is converted, presumably by a phosphatase, to B12. Mild heating of the bacteria appears to inactivate the phosphatase, and increases the yield of the precursor. A biosynthetic route is proposed, involving condensation of guanosine diphosphate cobinamide with α-ribazole 5'-phosphate. Two new compounds, α-ribazole 3',5'-diphosphate and a corrinoid containing a total of three phosphate groups, have been obtained by chemical synthesis.
