To examine the relationship between starvation-induced increases in UCP2 and UCP3 expression and mitochondrial proton conductance in rat skeletal muscle.
Rats were subjected to a 24-hour starvation intervention.
UCP2 and UCP3 mRNA and UCP3 protein expression were quantified using Northern and Western blot assays.
Mitochondrial proton conductance was measured by titrating respiration rate and membrane potential using malonate.
Starvation increased skeletal muscle UCP2 mRNA levels more than 5-fold, UCP3 mRNA levels more than 4-fold, and UCP3 protein levels by 2-fold.
Mitochondrial basal proton conductance remained unchanged despite the significant upregulation of UCP2 and UCP3.
