To quantify the proportion and absolute abundance of seven myosin heavy chain mRNA isoforms in rat muscles of divergent developmental origins.
Examined adult rat somatic limb muscles (extensor digitorum longus, tibialis anterior, soleus) and nonlimb branchial/head muscles (extraocular, thyroarytenoid, diaphragm, masseter).
Performed competitive PCR using an engineered exogenous competitor template containing primers specific for beta-actin and seven sarcomeric myosin heavy chain isoforms.
Soleus expressed primarily beta-cardiac MHC (87.8 ± 2.6%), while extraocular muscle was the only tissue to express all seven isoforms, including extraocular (7.4 ± 1.5%), neonatal (7.9 ± 0.3%), and embryonic (1.2 ± 0.5%) transcripts.
Absolute MHC mRNA levels varied from 64 pg/0.5 µg total RNA in tibialis anterior to 106 pg/0.5 µg total RNA in masseter, accounting for 4% to 17% of the total skeletal muscle mRNA pool.
An inverse expression relationship occurred between beta-cardiac and type 2B MHC transcripts across examined muscles, indicating reciprocal regulation.
