Abstract 1. The major classes of neutral lipids and the phospholipids were determined in sarcolemma, fragmented sarcoplasmic reticulum, and mitochondria of rat skeletal muscle. 2. The lipid composition of the sarcolemma is clearly different from that of sarcoplasmic reticulum and mitochondria. The sarcolemma contains a high content of phosphatidylserine and sphingomyelin in the phospholipid as well as large amounts of cholesterol, cholesterol ester and fatty acids in the neutral lipid fraction. 3. The fatty acid content of the neutral lipids is high in 16:0 + 18:0 and is less unsaturated than that of mitochondria or fragmented sarcoplasmic reticulum, which are very similar. Each of the cell fractions shows a distinct fatty acid pattern in the phospholipid fraction.
