To investigate the kinetics, isomer selectivity, and competitive interactions involved in carotenoid uptake and chylomicron secretion using intestinal cell models.
Cultured differentiated CaCo-2 cell monolayers on permeable membranes in the presence of oleate and taurocholate to stimulate chylomicron assembly and secretion.
Measured uptake and secretion of all-trans, 9-cis, and 13-cis β-carotene, α-carotene, lutein, and lycopene across time (2–16 h) and concentrations, comparing selectivity in HSC-T6, EAHY, U937 cells, and rat liver microsomes.
β-Carotene uptake and secretion into chylomicrons were saturable and concentration-dependent with an apparent Km of 7–10 µM; all-trans β-carotene absorption reached 11% (80% secreted into chylomicrons), whereas 9-cis and 13-cis isomers reached only 2–3%.
Absorption in CaCo-2 cells was 10% for α-carotene, 7% for lutein, and 2.5% for lycopene, with mutual competitive inhibition demonstrated between lycopene/β-carotene and β-carotene/α-carotene.
