Autotrophic cultures of E. gracilis contain a-tocopherol, -but other forms have not been dectected.1* While Threlfall and Goodwin2) reported the subcellular distribution and formation of the terpenoid quinones and related compounds, including a-tocopherol, in Euglena gracilis, the amount of a-tocopherol and its subcellular distribution have remained unknownowing to isolation of organelles by damaging ultrasonic treatment, incomplete separation of tocopherol using alumina columns, and spectrophotometric measurement of tocopherol based on the Emmerie-Engel reaction. In this paper we report on the separation of tocopherols by high-performance liquid chromatography (HPLC), the effects of illumination on the tocopherol content, and their subcellular distributions in E. gracilis z.
