Endothelin receptor antagonists and antibodies show potential therapeutic benefit in experimental models of various renal diseases.
Endothelin antagonists merit clinical trials in renal disease; evidence leaves open translation from experimental models.
Endothelin-1 (ET-1), the most potent vasoconstrictor yet identified, mediates a multitude of responses in various tissues including the kidney. The biological responses of ET-1 are mediated by specific cell surface receptors classified as ET(A) and ETB. Species differences are observed in the distribution as well as function of these ET receptors. 2. Involvement of ET has been demonstrated in a number of renal diseases, including ischaemia-induced acute renal failure, chronic renal failure, radiocontrast and cyclosporin-induced nephrotoxicity. ET antibodies as well as ET receptor antagonists have been shown to be beneficial in these disease models.
