To investigate the inhibitory role and cellular source of interleukin-4 (IL-4) against inflammatory mediator-induced mechanical hyperalgesia.
Evaluated mechanical hyperalgesia in rats after intraplantar injection of carrageenin, bradykinin, TNFalpha, IL-1beta, IL-8, or PGE2 with or without IL-4 pretreatment.
Measured IL-1beta and PGE2 production in LPS-stimulated murine peritoneal macrophages following 2-hour or 16-hour pre-incubation with murine IL-4.
Assessed mechanical nociceptive responses following anti-IL-4 antibody administration in control, athymic, and mast cell-depleted rats.
Pretreatment with IL-4 30 minutes prior blocked carrageenin-, bradykinin-, and TNFalpha-induced hyperalgesia, whereas IL-4 administered 12 or 12+2 hours prior inhibited IL-1beta hyperalgesia by 30% and 74%, respectively.
In LPS-stimulated murine macrophages, IL-4 pre-treatment for 2 hours suppressed IL-1beta production by 40%, whereas 16-hour pre-treatment inhibited PGE2 production rather than IL-1beta.
Neutralizing endogenous IL-4 with antibodies potentiated hyperalgesic responses in intact and athymic rats, but this potentiation was absent in mast cell-depleted rats.
