Physiologic TFPI may not regulate prothrombinase in vivo; challenges amidolytic assay interpretations and leaves open therapeutic targeting in animal models.
Contrary to results in amidolytic assays, prothrombinase is resistant to inhibition by physiological concentrations of TFPI in the presence of prothrombin, requiring much higher concentrations for effective blockade.
Mast et al. (1996) studied this question. Tissue factor pathway inhibitor (TFPI) was evaluated on Thrombin generation / prothrombinase inhibition. Physiologic concentrations of TFPI (< 8 nmol/L) minimally inhibit thrombin generation when prothrombinase is preformed, whereas higher concentrations (~100 nmol/L) effectively block its activity.
