The midpoint reduction potential (E m , pH 10) of the acceptor X was estimated in purified photosystem I particles prepared with Triton X-100, by reductive titration at gold electrode in the presence of bipyridyl dyes, and direct detection of the reduced acceptor by low temperature electron-spin resonance spectroscopy (ESR). The value obtained was —705 =±= 15 mV. The method of reduction offers advantages over chemical reduction techniques.
