A method was developed for the preparation of o~a-casein A from whole casein containing the mixed variant a~l-casein AB. It involved selective degradation of a~l-casein B by pepsin or rennet and isolation of a~l-casein A from the mixture of degradation products by DEAE-cellulose chromatography.
