The genus Pseudomonas (family Pseudomonadaceae) is among the most ecologically versatile bacterial genera, occupying soil, water, plant rhizospheres, and diverse host-associated environments, and characteristically encoding broad metabolic repertoires including siderophores, antibiotics, and secondary metabolites relevant to plant growth promotion and biocontrol [1].Strain Con_6 was recovered from agricultural soil in the Republic of Korea and provisionally identified as Pseudomonas sp. by 16S rRNA gene sequencing; wholegenome sequencing was undertaken to resolve its species identity and characterize its genomic features.Strain Con_6 was isolated from agricultural soil collected in Chilgok, Daegu, Republic of Korea (35°95′28.97″N,128°56′16.01″E).Soil was suspended in 0.85% (w/v) NaCl, serially diluted, and plated on tryptic soy agar (TSA; Difco, USA) at 37℃ for 24-48 h.A single colony was subcultured three times for purity and grown overnight in TSB at 37℃ and 200 rpm.Genomic DNA was extracted using the Wizard Genomic DNA Purification Kit (Promega, USA) with inversion mixing to preserve high-molecular-weight DNA for long-read sequencing.DNA quality was verified by NanoDrop and Qubit at the KNU NGS Core Facility (Republic of Korea).Whole-genome sequencing was performed on the Oxford Nanopore MinION platform using the Native Barcoding Kit V14 (SQK-NBD114-24) with an R10.4.1 flow cell.Read quality statistics summarized by NanoStat v1.6.0 yielded 38,514 reads (total 656.9 Mb; read length N50, 28,735 bp; mean quality, Q20.2; longest read, 169,787 bp).Low-quality reads and those shorter than 1,000 bp were removed with Filtlong v0.2.1, and the filtered reads were assembled de novo using Flye v2.9.5 [2] in --nano-hq mode with a target genome size of 6.5 Mb, with 3 rounds of internal polishing (-i 3); no additionalWe report the complete genome sequence of Pseudomonas sp.Con_6, a putative novel species isolated from agricultural soil in the Republic of Korea. The genome comprises a single circular chromosome of 7,034,194 bp (GC content, 63.43%).Phylogenomic analyses via ANI (91.945%) and dDDH (d4, 45.4%) against the closest type strain, Pseudomonas ulcerans, both fell below species demarcation thresholds, supporting novel species recognition.Functional annotation revealed 10 biosynthetic gene clusters including a pyoverdine siderophore locus, 99 carbohydrate-active enzymes, 4 antibiotic resistance genes, and 28 mobile genetic elements.
