GPER agonist LNS8801 reverses OVX-induced arterial stiffness in mice by reducing pulse wave velocity.
Why the study?
Arterial stiffness dramatically increases during menopause, but whether menopausal arterial stiffening resembles aging and whether GPER activation can reverse it was unknown.
Does GPER activation reverse arterial stiffness in a mouse model of menopause?
Population
Female C57Bl/6J mice ovariectomized at 10 weeks of age or aged to 52 weeks
Comparison
Ovariectomy vs aging, and GPER agonist treatment vs untreated
Design
Animal experimental study

GPER agonism reverses OVX-induced stiffness in mice; leaves open translation to human menopause.
Does GPER activation reverse arterial stiffness in a mouse model of menopause?
p-value: p=0.004
Ovariectomy and aging induce arterial stiffening via different mechanisms, and GPER activation with LNS8801 can reverse ovariectomy-induced stiffness in mice.
Kilanowski‐Doroh et al. (2023) studied Arterial stiffness. LNS8801 vs. Vehicle or G-1 was evaluated on Pulse wave velocity (PWV) (p=0.004). The enantiomerically pure GPER agonist LNS8801 significantly reduced pulse wave velocity to pre-ovariectomy levels (P=0.004), reversing ovariectomy-induced arterial stiffness.
