Establishes NO-mediated vasodilator tone in conscious rats; leaves open translation to human endothelial therapies.
In conscious rats, inhibition of NO synthesis with l-NMMA or l-NAME causes dose-dependent hypertension and regional vasoconstriction, highlighting substantial NO-mediated vasodilator tone in vivo.
Gardiner et al. (1991) studied this question. NG-monomethyl-l-arginine (l-NMMA) and NG-nitro-l-arginine methyl ester (l-NAME) was evaluated on Hemodynamic responses (blood pressure, heart rate, regional blood flows). Intravenous l-NMMA and l-NAME produced dose-dependent hypertension and marked regional vasoconstriction in conscious rats, consistent with substantial NO-mediated vasodilator tone.
