Key result
Disulfiram irreversibly suppresses protein synthesis and induces novel stress proteins in vascular endothelial cell lines.
Why the study?
The effects of the copper ionophore disulfiram on protein synthesis in vascular endothelial and hybridoma cells were not well characterized.
Does disulfiram alter protein synthesis in vascular endothelial and hybridoma cells?
Does disulfiram alter protein synthesis in vascular endothelial and hybridoma cells?
Disulfiram causes rapid and irreversible depression of key protein synthesis and induces stress proteins in vascular endothelial and immune cells in vitro.
Disulfiram may inhibit endothelial protein synthesis in vitro; hypothesis-generating and should not change practice.
Treatment of two clonal lines of vascular endothelial cells, two lines of antibody producing hybridoma cells and their parental line of myeloma cells with the anti-alcoholism copper lonophore, disulfiram, leads to rapid and irreversible depression of protein synthesis. In particular, synthesis of the biologically active molecules fibronectin, collagen and immunoglobulins was depressed. In contrast, disulfiram induced several novel ‘stress’ proteins in endothelial cells. These results suggest that clinical use of disulfiram may cause profound biochemical imbalance in tissues of the vasculature and of the immune system.

Hannan et al. (1982) studied this question. Disulfiram was evaluated on Protein synthesis. Treatment of vascular endothelial and hybridoma cell lines with disulfiram led to rapid and irreversible depression of protein synthesis, while inducing novel stress proteins in endothelial cells.
