Specific domains of sarcomeric alpha-actinin, including the actin-binding site and spectrin repeats, are dispensable for targeting the protein into Z bands in maturing myofibrils.
Lin et al. (1998) studied this question. Transfection with MYC-tagged s-alpha-actinin peptides was evaluated on Incorporation into Z bands and cytotoxicity. Transfection of myogenic cultures with MYC-tagged s-alpha-actinin peptides showed that the actin-binding site, spectrin-repeats, and C-terminal domains are dispensable for targeting into Z bands.
