AbstractAlthough skin diseases such as hyperpigmentation, aging, acne vulgaris etc are most commondermatological disease characterized by excessive melanin production from melanocytes cells itis resulting in uneven skin tone and dark patches and pigment, cosmetic concerns. MostConventional topical formulations such as creams and gels are widely used for its treatment. Butthey often exhibit limited skin penetration, poor stability of active ingredients. To overcome thisproblem we decided to formulate the nanostructured lipid carrier into a gel base. In recent years, transdermal drug delivery systems is most popular and promising drug delivery system whichprovide controlled and sustained delivery of therapeutic agents through the skin into the systemiccirculation at predetermine rate. which is further enhanced the potential of TDDS by improvingdrug solubility, stability, and skin permeation. Marula oil (natural fruits) and niacinamide(vitamin B3) is a very promising combination for the management of hyperpigmentation. Marulaoil is a natural lipid rich in antioxidants and essential fatty acids, anti- inflammatory effect whichhelp in skin barrier repair, moisturization, and protection against oxidative damage. Niacinamide(vitamin B3) is a well-established anti-pigmenting agent which is reduces hyperpigmentation byinhibiting melanin transfer. It is improving skin texture, and increases anti-inflammatoryproperties. The synergistic action of both agents enhances skin lightening, reduced inflammationwhile maintaining skin health and decrease dark spot and pigments. These formulation design, evaluation parameters (physiochemical properties, In vitro skin permeation study) andpreparation methods are hot high Pressure homogenization for NLC and incorporation methodsfor transdermal gel containing marula oil and niacinamide using NLCs. Overall, NLC-based transdermal gels offer a safe, effective, and patient-friendly approach for the dermatologicalapplication.
