Why the study?
Does the α1D-adrenergic receptor vaccine ADRQβ-004 mitigate heart failure with preserved ejection fraction (HFpEF) in a 'two-hit' HFpEF male ApoE mouse model?
Does the α1D-adrenergic receptor vaccine ADRQβ-004 mitigate heart failure with preserved ejection fraction (HFpEF) in a 'two-hit' HFpEF male ApoE mouse model?
In a preclinical mouse model, the novel α1D-adrenergic receptor vaccine ADRQβ-004 ameliorates HFpEF by modulating adipocyte-cardiomyocyte crosstalk via the PPARβ/AMPK/PGC1-α pathway.
May support α1D-AR vaccination as HFpEF therapy; hypothesis-generating in mice and leaves open human translation.
AIMS: Heart failure with preserved ejection fraction (HFpEF) remains a global clinical challenge with high mortality and restricted therapeutic options. Persistent sympathetic nervous system (SNS) overactivation, systemic metabolic dysregulation, and chronic inflammation are hallmarks of HFpEF pathogenesis. We developed ADRQβ-004, a novel therapeutic vaccine targeting α1D-adrenergic receptor (α1D-AR), to evaluate its potential in mitigating HFpEF. METHODS AND RESULTS: A "two-hit" HFpEF mouse model was established in 6-week-old male ApoE CONCLUSION: The ADRQβ-004 vaccine ameliorates HFpEF by modulating adipocyte-cardiomyocyte crosstalk through the PPARβ/AMPK/PGC1-α pathway. These findings suggest that ADRQβ-004 may serve as a promising immunomodulatory strategy for HFpEF research.

Wu et al. (2026) studied Heart failure with preserved ejection fraction (HFpEF). ADRQβ-004 (α1D-adrenergic receptor vaccine) was evaluated on Mitigation/amelioration of HFpEF. The ADRQβ-004 vaccine targeting the α1D-adrenergic receptor ameliorates HFpEF in mice via the PPARβ/AMPK/PGC1-α pathway (no quantitative effect estimates reported in abstract).
