Olenamidonin B is a cyclic lipopeptide antibiotic recently isolated from Streptomyces olivaceus. It exhibits antibacterial activity against Gram-positive bacteria and is especially potent against multidrug-resistant Enterococci. Its unusual bicyclic architecture, featuring a four-amino-acid macrocycle, an imidazolidinone ring, and a rare exocyclic (E)-dehydro-β-alanine-containing fatty acid, presents a compelling synthetic target. Here, we describe the total synthesis of olenimidonin B.
