The assembly of small molecules via multicomponent reactions represents a powerful strategy in organic synthesis. This study describes an efficient protocol for the synthesis of novel hexahydropyrimidine derivatives through a multi-component Quasi-Mannich condensation of 4-nitrobenzaldehyde, ammonium acetate, and nitroethane (in a 3:2:1 molar ratio) in n-butanol. The resulting structure served as versatile precursor for synthesizing various macromolecules and heterocyclic derivatives, starting with the selective reduction of two nitro groups into amino groups, followed by the synthesis of novel Schiff bases and corresponding seven-membered heterocyclic compounds. All the synthesized derivatives were comprehensively analyzed using advanced spectroscopic techniques, including Fourier-transform infrared spectroscopy (FT-IR), proton nuclear magnetic resonance (1H-NMR), and mass spectrometry. Furthermore, the target compounds were tested against Enterococcus faecalis using standard assays compared to conventional antibiotics such as beta-lactam antibiotics. The results showed that the biological activity depended on the effectiveness of the substituent groups in the compound
