In vitro study demonstrates effective drug delivery characteristics in controlled-release formulations, indicating potential for enhanced therapeutic effects.
Lercanidipine blocks the entry of extracellular calcium into vascular smooth muscle cells and cardiac muscle cells. Myocardial smooth muscle cells are unable to contract as a result of a drop in intracellular calcium, leading to a dilation of coronary and systemic arteries and a consequent reduction in blood pressure. Lubricated granules were compressed by rotary machine having round concave shaped punches with an average weight of 500 mg& min hardness of 5-6 kg/cm2. BD and the TD were found to be almost the same for all formulations. Both the CI and the HR were found to be between ? 18 and 1.09 and 1.21. This means that the blends flow well and can be compressed. All of the formulas had angles of repose between 22.17 and 31.11 degrees. It was found that tablets were between 5.82 and 5.91 mm thick. For different mixes, the hardness was found to be between 5.9 and 6.3 kg/cm2. All of the formulations had a friability of less than 1.0 %W/W. The amount of drug was found to be between 98 and 102%. The order of CR F9>f7>F8>from the dissolution data evident that the order of CR was It is evident that CR was better attained with combination of HPMC & the two natural polymers, then HPMC + single Natural polymer or HPMC alone.
