Introduction. Chronic wound healing in occupational dan non-occupational injuries are aesthetically, physiologically and psychologically disturbing. Traditional medicine like royal jelly has recognised to accelerate the healing process. There was no research has yet been done to identify royal jelly inggredients from Sragen, East Java. On this study intended to identify the isolates of RJ Sragen, East Java. Methods. This research is an original research paper and descriptive study. The research was conducted at Badan Riset dan Inovasi Nasional (BRIN) and Integrated Laboratory Research Center (ILRC). RJ was harvested at Griya Madu Salsabila Bee Farm Sragen, East Java in December 2021. RJ Quantitative examination is carried out to calculate the percentage (%) of flavonoids and phenols as well as qualitative liquid chromatography mass spectrometry (LCMS) to isolate/identify the contents of the active substance. Results. Seven isolates with prominent molecular mass (between 150-1200g/mol) were detected (phenols, flavonoids, adenosins, mahuanin G, candidate Mass C10H15N3O5, trans-4-hydroxy-3-methyl-6-(1-methiletyl)-2-cyclohensen-1, trans-dihydrocarbon and vitamin B5). Vitamins and other substances like amino acids were not detected, because the molecular weight of vitamins and amino acids is less than 150g/mol. Discussion. There are two RJ isolates that play a role in wound healing: 10-HDA and Defensin-1. Both were not detected in the LCMS examination. As an input for researchers, it is best to include the name of the type of isolate that they want to be tested, following the molecular mass rate. More research is needed to assess the effectiveness of RJ on human wound samples.
