Objectives: After the phenomenon of NAD+ resistance to aging was discovered, supplementing NAD+ has become a hot topic in anti-aging research. Among all the related ingredients, there have been quite a few preclinical and human clinical studies on NR that have been made public compared to other NAD+ candidates. In summary, NR, as a key NAD+ precursor, is helpful in increasing NAD+ levels and ameliorate effects of aging in animal models and clinical studies. But studies of its effects on improving naturally aged skin condition are limited. The study aim is to evaluate the skin benefits through a clinical study of a certain formula which was built up with grape seed extract, vitamin C, vitamin E and rosehip extract besides NR. Methods: Effects of the NR contained formula were investigated in a 28-day randomized clinical trial involving 35 Asian female participants aged 30-45, which are all had related facial skin problems as evaluated by dermatologists. Indicators of skin conditions including moisture, whitening, brightening, facial spot-fading, firmness, smoothness and facial anti-wrinkle effect were measured during the study. And a questionnaire including questions about the skin recovery, daily energy and mood status were taken. We objectively measured these indicators every other week. Statistical analysis was performed using the t test. Values of p<0.05 were considered statistically significant. Results: Remarkably, administered NR with grape seed extract, vitamins and rosehip extract could significantly improve skin condition, including moisture, whitening, brightening, spot-fading and firmness after 14 days (p<0.001). Furthermore, besides these indicators, skin smoothness and anti-wrinkle could be significant improved after 28 days consumption(p<0.001). The questionnaire indicated that more than 88% participants felt skin recovery, daily energy and mood status all got improved. Conclusions: In summary, these results showed NR contained formula with grape seed extract, vitamins and rosehip extract could improve skin-condition of naturally aged Asian women.
