In vitro toxicity curves. LS174T cells were seeded in 96 well plates and then incubated with a serial dilution of either LG or XAb-SN-38 for 6 days with media replacement every 24 hours. The toxicity curves are presented in either protein (A) or payload (B) incubation concentration. Although XAb-SN-38 provided more efficacy in vivo, the two ADCs resulted in similar in vitro potency.
