Key Points
- To determine whether atrial natriuretic factor inhibits the aldosterone response by altering calcium influx and efflux stimulated by angiotensin II or potassium in rat adrenal glomerulosa cells.
- Isolated rat adrenal glomerulosa cells were analyzed using a superfusion system.
- Cells were stimulated with potassium ions (8 mM or 12 mM) or angiotensin II (10^-9 M and 10^-8 M) in the presence or absence of atrial natriuretic factor (10^-8 M) to evaluate calcium flux rates.
- Potassium (8 mM) significantly elevated Ca2+ influx compared to basal levels (0.91 ± 0.10 vs 0.42 ± 0.04 nmol/min/10^6 cells; p < 0.01), but atrial natriuretic factor did not inhibit this response (0.99 ± 0.18 nmol/min/10^6 cells).
- Angiotensin II stimulated Ca2+ influx (10^-9 M: 0.62 ± 0.02, 10^-8 M: 0.71 ± 0.09 vs basal: 0.44 ± 0.03 nmol/min/10^6 cells; p < 0.05), and atrial natriuretic factor produced no inhibitory change on this influx (0.62 ± 0.06 and 0.69 ± 0.14 nmol/min/10^6 cells, respectively).
