Demonstrates effective regeneration of Achnatherum splendens, highlighting viable cultivation methods.
We studied the regeneration of Achnatherum splendens (Trin.) Nevski in vitro and selected several favorable media. Germ-free plants were produced with germination on MS medium. Callus was induced with B5+1.5 mg·L⁻¹ 2,4-D+0.5 mg·L⁻¹ NAA, with leaf sheath and hypocotyls as explants. After two subcultures, shoots and roots were induced. The medium containing B5+ 0.5 mg·L⁻¹6-BA + 0.2 mg·L⁻¹ NAA was suitable for shoot induction. For root induction, the medium containing 1/4 B5+1.0 mg·L⁻¹ NAA+ 0.2 mg·L⁻¹ IBA +1.0 g·L⁻¹ active carbon was favorable. With this regeneration system for A. splendens (Trin.) Nevski, we achieved the highest survival rate of transplants.
