Are specific gene single nucleotide polymorphisms associated with the presence of hypertension in patients with obstructive sleep apnea?
p-value: p=<0.05
Variations in the rs12511115 SNP of the GABRB1 gene may be associated with the development of hypertension in patients with obstructive sleep apnea.
Identifies a potential genetic marker for hypertension risk in obstructive sleep apnea; leaves open whether GABRB.
Objective To further explore the differences in nucleotide sequences of 20 gene loci closely related to hypertension in patients with obstructive sleep apnea (OSA), we conducted a study by detecting these loci in patients with and without hypertension. Subjects and Methods All patients were from 319 cases of OSA patients with and without hypertension who were hospitalized in the respiratory sleep department of Karamay central hospital from January 2023 to September 2024. Blood samples were collected from all patients, The patients included 220 cases of OSA with hypertension, with an average age of (56.6 ± 11.5) years. There were 114 males and 106 females. There were 99 cases of OSA without hypertension, with an average age of (48.5 ± 11.0) years. and 100 cases hypertension without OSA individuals from the same period were included as the control group. including 90 males and 10 females, with an average age of (50.6 ± 7.4) years. The genomic DNA was extracted using a Wizard Genomic DNA Purification Kit (Promega, Madison,Wisconsin, USA) as per the product instruction. A multiplex PCR-ligase detection reaction method was used for genotyping four SNPs. For each SNP, the alleles were distinguished by different fluorescent labels of allelespecific oligonucleotide probe pairs. Different SNPs were further distinguished by different extended lengths at 3’end. Results By comparing the 20 gene loci closely related to OSA, including rs10256629, rs10423928, rs10811640, rs10917318, rs11030323, rs1137101, rs1143633, rs116618654, rs117228173, rs11831172, rs12511115, rs1327235, rs140929616, rs1409986, rs144468328, rs17372813, rs3918242, rs6313, rs731236, and rs7975232, between the groups (OSA with hypertension group, OSA without hypertension group), it was found that the C/G (SNP)in rs12511115 of the gamma-aminobutyric acid (GABA) A receptor GABRB1 gene on chromosome 4 between the OSA with and without hypertension groups (P < 0.05). Conclusion The variations of the C/G SNP in rs12511115 of the GABRB1 gene may be related to OSA with or without hypertension in different ethnic groups. The related mechanisms need further study. H Zhongming (2026) conducted an observational in Obstructive Sleep Apnea (OSA) complicated with hypertension (n=419). Genotyping of 20 gene loci vs. OSA without hypertension and hypertension without OSA was evaluated on Differences in nucleotide sequences of 20 gene loci (p=<0.05). The C/G single nucleotide polymorphism in rs12511115 of the GABRB1 gene was significantly different between obstructive sleep apnea patients with and without hypertension (P < 0.05).
