Population
Male rats in a model of large myocardial infarction
Design
Preclinical, randomized to proximal left anterior descending ligation
Follow-up
up to 35 days
Key result
In a rat model of myocardial infarction, TNF-alpha protein levels rose 8- to 10-fold above normal in the infarct and peri-infarct zones and 4- to 5-fold in the contralateral zone.

TNF-α expression beyond infarct zones broadens post-MI remodeling targets; leaves open human therapeutic relevance.

TNF-alpha production forms an important intrinsic myocardial stress response to injury, with expression occurring early and sustained in both infarcted and non-infarcted myocardium.
Irwin et al. (1999) conducted an RCT in myocardial infarction. proximal left anterior descending ligation vs. normal levels was evaluated on gene expression and protein production of TNF-alpha and TNF-R1/R2. In a rat model of myocardial infarction, TNF-alpha protein levels rose 8- to 10-fold above normal in the infarct and peri-infarct zones and 4- to 5-fold in the contralateral zone.
