In vivo, the positive inotropic effect of ET-1 is masked by ETA-mediated coronary vasoconstriction and ischemia, but can be unmasked by pretreatment with molsidomine or an ETA receptor antagonist.
Beyer et al. (1999) studied this question. Endothelin-1 (ET-1) with or without molsidomine or BQ 610 vs. Saline controls was evaluated on Cardiac output, total peripheral resistance, and isovolumic peak first derivative of left ventricular pressure.
