Structured PICO

Does silencing circ_0124644 alleviate hypoxia-induced cardiomyocyte injury in an in vitro model of acute myocardial infarction?

P
Population
31 patients with acute myocardial infarction (AMI) diagnosed by angiography (excluding severe liver and kidney dysfunction, blood system diseases, cancer, and infectious diseases) and 25 healthy control volunteers. In vitro model: Human cardiomyocytes (AC16) cultured under hypoxia (1% O2).
I
Intervention
In vitro: Hypoxia treatment (1% O2 for 24 h), lentivirus short hairpin RNA targeting circ_0124644 (sh-circ_0124644), miR-590-3p mimic/inhibitor, and pcDNA SOX4 overexpression vector.
C
Comparator
In vitro: Normoxia (21% O2), control shRNA (sh-NC), control miRNA (miR-NC, anti-NC), and empty pcDNA vector. In vivo: Healthy control volunteers.
O
Outcome
Expression levels of circ_0124644, miR-590-3p, and SOX4; cell viability, cell cycle progression, apoptosis, and oxidative stress markers (LDH, MDA, SOD, CAT).surrogate

Circ_0124644 silencing alleviates hypoxia-induced cardiomyocyte injury by regulating the miR-590-3p/SOX4 axis, presenting a potential therapeutic target for acute myocardial infarction.
