Implication

High-affinity dyes may overestimate Cai2+ durations in animal myocardium; leaves open optimal dye selection for accurate calcium imaging studies.

Structured PICO

P
Population
Porcine left ventricular (LV) wedge preparations (n=13)
I
Intervention
Local intramural injection of high-affinity Ca2+-sensitive dyes (Fluo-4, Fluo-2MA, or Rhod-2)
C
Comparator
Local intramural injection of low-affinity Ca2+-sensitive dyes (Fluo-4FF or Fluo-2LA)
O
Outcome
Duration (CaD50, CaD80) and shape of intracellular calcium (Cai2+) transients at various pacing cycle lengthssurrogate

High-affinity calcium-sensitive dyes significantly overestimate the duration and alter the shape of intracellular calcium transients in intact ventricular myocardium compared to low-affinity dyes.

Limitations

  • Dye Kd values were obtained in test solutions and may be substantially different inside cells
  • Microcapillary staining did not work every time, yielding usable signals in only ~70% of injections
