Overview

May indicate genetic risk for methylphenidate CV effects in ADHD; hypothesis-generating and requires validation before any clinical application.

Structured PICO

Are norepinephrine transporter and alpha-2A-adrenergic receptor polymorphisms associated with cardiovascular side effects of methylphenidate in children with ADHD?

P
Population
101 drug-naive Korean children with attention-deficit/hyperactivity disorder (ADHD), mean age 8.7 ± 1.7 years.
I
Intervention
Osmotic Controlled-Release Oral Delivery System-methylphenidate (OROS-MPH) titrated based on symptom severity and side effects for 12 weeks.
O
Outcome
Percentage change in resting heart rate (HR), diastolic blood pressure (DBP), and systolic blood pressure (SBP) before and after 12 weeks of treatment.safety

Norepinephrine-related gene polymorphisms (SLC6A2 and ADRA2A) are associated with significant increases in heart rate and diastolic blood pressure in children treated with methylphenidate for ADHD.
