The preparation of derivatives of aldosterone for use in GLC separations was studied. Several derivatives with good gas chromatographic properties were prepared. Since this steroid hormone is usually present in biologic systems in very low concentration, the chief requirement is for a derivative or derivatives that may be used with an electron capture detector. The HFB-MO derivatives (four isomers) separate as a single peak with a non-selective column, and these derivatives may be detected at nanogram and subnanogram levels. The structures of the derivatives prepared in this study were determined by mass spectrometry. Aldosterone reacts primarily as the C-18 hemiacetal although a tri-MO structure may be prepared by prolonged reaction with methoxylamine. Derivatives of the hemiacetal structure have the greatest usefulness for GLC work.
