Acetylcholine-induced relaxation was significantly reduced in SHRSP resistance arteries compared to WKY controls (61 ± 5%, IC50 > 6.6 ± 0.4, n = 24 vs. 88 ± 2%, IC50 7.3 ± 0.1, n = 31; P < 0.005).
Relaxations to exogenous nitric oxide remained preserved in SHRSP, and treatment with the cyclooxygenase inhibitor meclofenamate fully restored acetylcholine-induced relaxation, whereas the thromboxane synthetase inhibitor CGS 13080 had no effect.
Relaxations to sodium nitroprusside were heightened in SHRSP both in arterial rings with and without endothelium.
