Renal cortical mRNA expression of ET-1, ET(A), ET(B), TGF-beta, TNF-alpha, and collagen type IV was quantified by RT-PCR, ET(B) protein was analyzed via Western blot, and interstitial expansion was measured by point counting.
Enalapril reduced ET-1 mRNA expression by 44% (P < 0.02) and increased ET(B) mRNA expression by 115% (P < 0.05) along with ET(B) protein in obstructed kidneys, but did not alter elevated ET(A) mRNA levels.
Enalapril alone decreased UUO-induced interstitial volume expansion by 52%, whereas co-administration with BQ-788 suppressed interstitial volume expansion by only 19%.
